Objectives:The goal of this study was to compare the effects of orlistat on weight gain in overweight and obese adults with orlistat use. Methods: We conducted a 1-period, randomized, parallel-group trial that enrolled 1,063 overweight and obese adults, divided into two groups: orlistat and an dummy treatment group. The primary outcome measure was body mass index (BMI) at baseline (1 month) and at each follow-up visit. Secondary outcomes included weight gain and changes in BMI over the follow-up period. Results: Weight gain was significantly greater in the orlistat group compared with orlistat group, with a significant difference between orlistat and the dummy group in all subgroups. Body mass index (BMI) at baseline was higher in the orlistat group than in the orlistat and dummy groups, whereas BMI at follow-up was higher in the dummy groups compared with orlistat. The results were consistent among subgroups, with differences in the change in BMI between the groups for orlistat and the dummy groups being greater than for orlistat. The change in BMI over follow-up was greater in orlistat than in the dummy groups. Conclusions: Orlistat and orlistat may have beneficial effects on weight gain in overweight and obese adults with orlistat use. Future research is needed to assess their long-term clinical efficacy in the long term.
Introduction to orlistat, orlistat-based weight loss medicationsOverview:Orlistat, a lipase inhibitor, is widely used to manage obesity and is also considered an effective weight loss treatment. It was first approved by the Food and Drug Administration (FDA) in 1999, and has been shown to be effective in numerous clinical trials. Orlistat is a lipase inhibitor that inhibits the production of dietary fat, reducing dietary fat absorption by inhibiting the lipase enzymes. A total of 1,063 participants were randomized to either orlistat or dummy treatment, and the study was stopped early after a median of 1.5 (range: 1.0-2.0) baseline weight. Participants who received orlistat had a BMI ≥25 kg/m2 or BMI ≥30 kg/m2 on baseline and at follow-up, and participants who received orlistat had a BMI ≥30 kg/m2 or BMI ≥30 kg/m2 at follow-up were eligible to participate. Of the 1,063 participants who received orlistat, 6.8% were treated with orlistat compared with 7.6% of participants who received placebo. In orlistat-treated participants, 5.3% lost at least 1 year of weight gain and 5.7% lost at least 1 year of weight loss compared with 7.6% of participants who received placebo. In orlistat-treated participants, the mean change from baseline was 0.6 kg in body weight, and the mean change from baseline was 0.4 kg in body weight in orlistat-treated participants. In orlistat-treated participants, the mean change from baseline was 0.9 kg in body weight, and the mean change from baseline was 0.8 kg in body weight in orlistat-treated participants. In orlistat-treated participants, the mean change from baseline was 0.8 kg, and the mean change from baseline was 0.9 kg in body weight, respectively. There was no difference in body weight gain between the orlistat and orlistat groups. Conclusion: Orlistat, a lipase inhibitor, has potential benefits in weight loss and may be used in conjunction with a BMI ≥25 kg/m2 or BMI ≥30 kg/m2 in overweight and obese adults with orlistat use.
